| I. Background | |
| GHSR1a,全稱為生長(zhǎng)激素促分泌素受體1a型,是功能完整的7次跨膜受體的G蛋白 偶聯(lián)受體,負(fù)責(zé)所有已知的胃饑餓素生物學(xué)效應(yīng),包括刺激食欲、促進(jìn)生長(zhǎng)激素釋放、調(diào) 節(jié)能量平衡和糖脂代謝等。GHSR1a以內(nèi)源性激素胃饑餓素為主要配體。它通過(guò)偶聯(lián)Gq/11 蛋白,核心信號(hào)通路為PLC-β,產(chǎn)生IP3并動(dòng)員細(xì)胞內(nèi)鈣離子,進(jìn)而觸發(fā)下游級(jí)聯(lián)反應(yīng); 此外,它還能在不同細(xì)胞環(huán)境中與Gi/o或Gs等通路交互,形成一個(gè)復(fù)雜的信號(hào)轉(zhuǎn)導(dǎo)網(wǎng) 絡(luò)。該受體廣泛分布于下丘腦、海馬體、胰腺及心臟等多組織,其信號(hào)精確調(diào)控著生長(zhǎng)激 素釋放、食欲、能量代謝及神經(jīng)保護(hù)等多種關(guān)鍵生理功能,從而成為治療惡病質(zhì)、肥胖及 相關(guān)代謝與神經(jīng)疾病的重要藥物靶點(diǎn)。 | |
